I prefer all of my pedals in line but especially overdrives. Sounds better that way. I run a Fender Twin Reverb '65RI which doesn't have an FX Loop but I've had amps with them and I've never cared for loops. I prefer everything in front of the amp. Even delays. It sounds more natural to me.

I use the W.H. Pork Loin inline, and I use a MXR 10 Band EQ thru my FXS Loop, which allows me to
tailor the freq response between the Pre-Amp and the Final amp. I also run my delay and reverb
thru the effects loop. I'm not partial to running them thru the front end with distortion devices,
because when I've done this in that past, I found the results to be very unfavorable.
I utilize this set up currently, on my Crate V32. I have a Marshall MV 50W 2187, and a Fender HRD3 that I
also utilize the effects loops on, with delay end reverb,, but no external EQ as yet. My plan is to acquire
additional MXR 10 band systems, and add them into the FXS Loops on those amps too. Again, I run all
distortion devices thru the front end only, Running the MXR 10 Band Q in the FXS Loop
gives you a HUGE amount of control over your signal, then your only concern is:
How good are your speakers, and are they giving you the kind of Freq response that you desire?

Always run your OD inline you want them to push the Pre-amp tubes to break up. And most OD don't sound very good in the loop. However an EQ in the loop can do a lot of cool things like create a clean boost or give you the right lead tone bump in all the right places.
